Exploring the Link Between Proton Pump Inhibitors and Dementia Risk

Introduction:

Recent research led by Nelsan Pourhadi, MD, from the Danish Dementia Research Centre, Copenhagen University Hospital–Rigshospitalet, has shed light on the potential link between PPIs and an increased risk of dementia. The study utilized Danish registries to analyze data from 1,983,785 individuals aged 60-75 years, focusing on dementia diagnoses and PPI use between 2000 and 2018.

Key Findings:

  • Individuals with a history of PPI use showed a significantly higher risk of developing dementia before age 90.

  • The highest risk was observed in those diagnosed with dementia before age 70.

  • The study recorded 99,384 cases of all-cause dementia, with a median age of diagnosis at 79 years.

  • Among younger individuals aged 60-69, the risk for dementia associated with PPI use was 36% higher.

  • This risk decreased to 6% in those aged 80-89.

  • No significant link was found between PPI use and dementia diagnoses after age 90.

  • The strongest association was noted among the youngest cases of dementia, indicating a possible critical exposure window in midlife.

  • While this study is significant, it's important to note its limitations, such as the lack of data on PPI prescriptions before 1995, over the counter PPI use, and in-hospital intravenous PPI use during the study period.
